Fluoro- or trifluoromethyl-substituted benzyl and phenethyl alcohols: Substrates for metal-mediated site-selective functionalization

It was possible to functionalize the three fluorobenzyl alcohols and the three 2-(fluorophenyl)ethanols by metalation and subsequent carboxylation, the prototype electrophilic trapping reaction. Triisopropylsilyl (TIPS) outperformed methoxymethyl (MOM) as an O-protective group making seven new fluorobenzoic acids accessible in 63% average yield. Moreover, the TIPS group tolerates weakly basic and acidic media and, therefore, may facilitate further structural elaboration. The unprotected alcohols reacted more sluggishly and were unable to provide two of the targeted products (acids 1 and 2). The yield averaged only 46% in the five other cases (acids 3-7). The direct metalation of fluorinated benzyl and phenethyl alcohols remains nevertheless an attractive option because of its operational simplicity. All three (trifluoromethyl)benzyl alcohols and two of the three (trifluoromethyl)phenethyl alcohol isomers were successfully submitted to the metalation/functionalization sequence. These five starting materials gave rise to a total of nine new benzoic acids or lactones (compounds 8-14 and 17-18). Despite the poor yields (31% on average), the organometallic methods employed are, in general, extremely selective, economical and easy to perform. Conformations and laser-induced fluorescence spectroscopy of jet-cooled 2-(p-fluorophenyl)ethanol

Vibronically-resolved S1-S0 electronic spectra of 2-(p-fluorophenyl)ethanol have been studied by use of laser-induced fluorescence excitation and dispersed fluorescence spectroscopic methods in a supersonic free jet expansion. Two conformational isomers of the molecule are identified, and with the aid of the predictions of quantum chemistry calculations these are assigned to gauche and anti conformers. The assignments are corroborated by analyzing the vibronic features in the dispersed fluorescence spectra of the two conformers and their TDDFT predicted electronic transition energies. Energetic and spectral features of the conformers are compared with those of the parent molecule 2-phenylethanol.

Antiproliferative, antiandrogenic and cytotoxic effects of novel caffeic acid derivatives in LNCaP human androgen-dependent prostate cancer cells

Caffeic acid and its naturally occurring derivative caffeic acid phenethyl ester (CAPE) have antiproliferative and cytotoxic properties in a variety of cancer cell lines without displaying significant toxicity toward healthy cells, and are considered to be potential anticancer agents. However, little is known about their effects on prostate cancer cells. We synthesized and evaluated the effects of caffeic acid, CAPE (2) and 18 synthetic derivatives on cell viability and androgen-dependent cell proliferation, subcellular localisation and expression of androgen receptor (AR) and secretion of prostate-specific antigen (PSA) in LNCaP human hormone-dependent prostate cancer cells. Several synthetic derivatives of CAPE were strong, concentration-dependent cytotoxic agents in LNCaP cells with IC50 values in the 6.8-26.6 muM range, potencies that were up to five-fold greater than that of CAPE (33.7 ¡À 4.0 muM). A number of caffeic acid derivatives were inhibitors of androgen-stimulated LNCaP cell proliferation with concomitant inhibition of DHT-stimulated PSA secretion. Compound 24 was the most cytotoxic and antiproliferative caffeic acid derivative (IC50 values of 6.8 ¡À 0.3 and 2.4 ¡À 0.8 muM, respectively) inhibiting DHT-stimulated cell proliferation and PSA secretion statistically significantly at concentrations as low as 0.3 muM. Exposure to DHT increased cytoplasmic and nuclear AR levels and co-treatment with increasing concentrations of compound 24 or CAPE (2), notably, further increased these levels. In conclusion, a number of synthetic derivatives of caffeic acid are potent inhibitors of androgen-dependent prostate cancer cell proliferation and viability, acting, at least in part, via an antiandrogenic mechanism that involves increased nuclear accumulation of (presumably inactive) AR.

Design, synthesis and biological evaluation of novel human monoamine oxidase B inhibitors based on a fragment in an X-ray crystal structure

Herein we report our efforts of developing reversible selective hMAO-B inhibitors based on isatin, a fragment in an X-ray crystal structure. Five different scaffolds were designed and many compounds were synthesized. Among them, compound A3 demonstrated very high potency and isoform selectivity against hMAO-B, 11 and 13 times more potent (IC50 = 3 nM) and 23.64 and 6.8 times more selective than the marked drugs, selegiline and safinamide. However, the endeavors to modify the polar 3-one group of isatin, that is in a hydrophobic environment in the binding site of hMAO-B, to small nonpolar hydrophobic groups did not bring about improved hMAO-B inhibitors, which may challenge our understanding of molecular interactions and molecular recognition in biological systems.

Anti-biofilm effect of novel thiazole acid analogs against Pseudomonas aeruginosa through IQS pathways

IQS has been proven to be a new quorum sensing (QS) system against bacterial biofilm formation, which is activated in the common phosphate-limiting environment of infected tissues taking over the central las system. Up to now, numerous biofilm inhibitors which function by affecting traditional QS system have been reported. However, no compound has been reported to exert anti-biofilm activity through IQS system. Herein, various novel IQS derivatives were synthesized by the reaction of thiazole-4-carboxylic acid with different linear alcohols (R-OH) or amines (R-NH2). IQS derivatives with four carbon chain length of R group were found to present the best biofilm inhibition activity. Compound B-11 as the model molecule was observed to inhibit biofilm formation only under phosphate-limiting condition, and increase in B-11 concentration significantly reduced the expression of rhlA-gfp and pqsA-gfp, but lasB-gfp. Moreover, B-11 reduced production of virulence factors of rhamnolipid and pyocyanin under phosphate limitation. These observations indicated that the synthesized compounds possessed the anti-biofilm activity through IQS pathways rather than traditional QS pathways, which pave a path for future molecular design against bacterial biofilm formation.

Stereo-selective bio-reduction of acetophenone and its derivatives by soil fungal isolates

Microbial asymmetric reduction of ketone is an efficient tool for the synthesis of chiral alcohols. Many reports are available for the bio-reduction of acetophenone and its derivatives, since it is widely used model substrate. This research focus on exploring the soil fungal isolates for their ability towards the reduction of acetophenone and its derivatives to their corresponding chiral alcohols using growing cells instead of resting cells. Bio-reduction of acetophenone, 4-fluoro acetophenone, and 4-chloro acetophenone were carried out using different fungal cultures isolated from soil. All the isolates exhibited a good reduction capability when grown in an optimal condition. Among the screened fungal cultures, Penicillium sp. and Aspergillus sp. showed significant bioconversion with varying enantio-selectivity. However, the Penicillium sp. has showed maximum ability of bio-reduction. The best performing isolate was characterized using internal transcribed spacer (ITS) region and found to be Penicillium rubens VIT SS1, which showed higher conversion and selectivity more than 90% towards acetophenone and its derivatives. The reaction conditions such as pH, temperature and media were evaluated for the bio-reduction of acetophenone using Penicillium rubens VIT SS1. The substrate loading was increased from 0.5g/L to 6g/L at shake flask level using the optimized condition pH 5¡À0.5 and temperature 25¡À2C. This study revealed huge potential of fungal cultures for the synthesis of many aromatic chiral alcohols in a simpler, novel and cost effective manner.
