Ferredoxin:NADP(+) oxidoreductase from Plasmodium falciparum (PfFNR) catalyzes the NADPH-dependent reduction of ferredoxin (PfFd), which provides redox equivalents for the biosynthesis of isoprenoids and fatty acids in the apicoplast. Like other flavin-dependent electrontransferases, PfFNR is a potential source of free radicals of quinones and other redox cycling compounds. We report here a kinetic study of the reduction of quinones, nitroaromatic compounds and aromatic N-oxides by PfFNR. We show that all these groups of compounds are reduced in a single-electron pathway, their reactivity increasing with the increase in their single-electron reduction midpoint potential (E-7(1)). The reactivity of nitroaromatics is lower than that of quinones and aromatic N-oxides, which is in line with the differences in their electron self-exchange rate constants. Quinone reduction proceeds via a ping-pong mechanism. During the reoxidation of reduced FAD by quinones, the oxidation of FADH(.) to FAD is the possible rate-limiting step. The calculated electron transfer distances in the reaction of PfFNR with various electron acceptors are similar to those of Anabaena FNR, thus demonstrating their similar intrinsic reactivity. Ferredoxin stimulated quinone- and nitro-reductase reactions of PfFNR, evidently providing an additional reduction pathway via reduced PfFd. Based on the available data, PfFNR and possibly PfFd may play a central role in the reductive activation of quinones, nitroaromatics and aromatic N-oxides in P. falciparum, contributing to their antiplasmodial action.

Leveraging the elevated levels of hydrogen peroxide (H2O2) in cancer, inflammatory diseases and cardiovascular disorders, H2O2-activated promoieties have been widely used in drugs and biomaterials design. However, the overwhelming majority of the promoieties only share the common structure of a H2O2-responsive arylboronic acid/ester moiety with low diversity. We report here an unprecedented strategy to construct novel H2O2-responsive prodrugs based on an alpha-ketoamide structure. As a proof of concept, we designed and synthesized a panel of alpha-ketoamide based nitrogen mustard prodrugs, among which KAM-2 showed potent growth inhibitory activity and high selectivity toward cancer cells. The H2O2-trigged decomposition of KAM-2 was validated, and the DNA damaging and apoptosis promoting activity attributed to the released nitrogen mustard were demonstrated. Our work unveils alpha-ketoamide as a new scaffold for prodrug design and may quickly inspire future developments.

Imidazo[1,2-a]pyridines form versatile scaffolds in pharmaceutical industry arising from their diverse biological activities. The synthesis of these molecules thus has been of great interest and resulted in the development of a large number of new methodologies. Herein we describe the first iron-catalyzed Ortoleva-King type protocol towards the synthesis of these fused heterocyclic compounds. This methodology employs cheap and easily available FeCl3 center dot 6H(2)O and molecular iodine as the catalytic system. The procedure has been well explored by extending the substrate scope with a variety of aromatic ketones and 2-aminopyridines to furnish different imidazo[1,2-a]pyridine derivatives in moderate to good yields. While dual photocatalysis-transition metal catalysis strategies are extensively reported, the majority of systems feature two separate catalysts, limiting the potential for synergistic interactions between the catalytic centres. In this work we synthesised a series of tethered dual catalysts allowing us to investigate this underexplored area of dual catalysis. In particular, Ir(i) or Ir(iii) complexes were tethered to a BODIPY photocatalyst through different tethering modes. Extensive characterisation, including transient absorption spectroscopy, cyclic voltammetry and X-ray absorption spectroscopy, suggest that there are synergistic interactions between the catalysts. The tethered dual catalysts were more effective at promoting photocatalytic oxidation and Ir-catalysed dihydroalkoxylation, relative to the un-tethered species, highlighting that increases in both photocatalysis and Ir catalysis can be achieved. The potential of these catalysts was further demonstrated through novel sequential reactivity, and through switchable reactivity that is controlled by external stimuli (heat or light).

A series of robust metallo-aerogels are readily fabricated by pyrolysis of xerogels derived from chitosan-metal (M = Fe, Co, Ni) hydrogels. Owing to the strong coordination between metal ions and the functional groups (NH2 and OH) of chitosan, metallo-aerogels consisting of encapsulated metal-nanoparticles (MNPs) by graphite shells were obtained, as supported by various characterizations including high-resolution transmission electron microscope (HR-TEM), X-ray diffraction (XRD), and Raman. The resulting metallo-aerogels could be functioned as highly stable, efficient and selective nanocatalysts towards the hydrogenation of nitroarenes to amines at low catalyst loading (1.2 mol.%-2.4 mol.%). Remarkably, the metallo-aerogels could be reused for more than 30 runs without obvious loss of activity and selectivity. These distinguished performances were attributed to the graphitic shells formed during the pyrolysis, which hampered the possible aggregation of MNPs, prevented metal leaching and increased their stability.

The reaction of coordinatively unsaturated Co(II) PCP pincer complexes with nitric oxide leads to the formation of new, air-stable, diamagnetic mono nitrosyl compounds. The synthesis and characterization of five- and four-coordinate Co(III) and Co(I) nitrosyl pincer complexes based on three different ligand scaffolds is described. Passing NO through a solution of [Co(PCPNMe-iPr)Cl], [Co(PCPO-iPr)Cl] or [Co(PCPCH2-iPr)Br] led to the formation of the low-spin complex [Co(PCP-iPr)(NO)-X] with a strongly bent NO ligand. Treatment of the latter species with (X = CI, Br) AgBF4 led to chloride abstraction and formation of cationic square-planar Co(I) complexes of the type [Co(PCP-iPr)(NO)](+) featuring a linear NO group. This reaction could be viewed as a formal two electron reduction of the metal center by the NO radical from Co(III) to Co(I), if NO is counted as NO+. Hence, these systems can be described as {CoNO}(8) according to the Enemark-Feltham convention. X-ray structures, spectroscopic and electrochemical data of all nitrosyl complexes are presented. Preliminary studies show that [Co(PCPNMe-iPr)(NO)](+) catalyzes efficiently the reductive hydroboration of nitriles with pinacolborane (HBpin) forming an intermediate {CoNO}(8) hydride species.

An efficient metal-free approach has been devised for the synthesis of novel beta-carboline C-1 tethered 2,3-dihydropyrroles and pyrrolesviaone-pot assembly involving 1-formyl beta-carbolines, aryl methyl ketones, and isonitriles. The reaction advances through the formation of beta-carboline linked enones followed by [3+2] cycloaddition with isonitriles. This methodology features an operationally simple procedure, multicomponent character, high atom economy, a broad substrate scope, short reaction times, and high regioselectivity with excellent yields. The scope of the process was exemplifiedviathe synthesis of a library of 34 novel compounds with significant diversity.

Ras proteins are small GTPases which regulate cellular proliferation, differentiation, and apoptosis. Constitutively active mutant Ras are expressed in similar to 15-20% human cancers, and K-Ras mutations account for similar to 85% of all Ras mutations. Despite the significance of Ras proteins in refractory cancers, there is no anti-Ras drug available in clinic. Since K-Ras must interact with the plasma membrane (PM) for biological activity, inhibition of the K-Ras/PM interaction is a tractable approach to block oncogenic K-Ras activity. Here, we discovered chalcones 1 and 8 exhibit anti-K-Ras activity, and show that the compounds mislocalize K-Ras from the PM and block oncogenic K-Ras signal output. Also, 1 inhibits the growth of K-Ras-driven human cancer cells. Our data suggest that 1 could be a promising starting point for developing anti-K-Ras cancer drug.
