A green and efficient visible-light promoted aerobic oxidative C-C bond cleavage of vicinal diols in micellar medium has been developed. This protocol used graphitic carbon nitride with nitrogen vacancies (CN620) as a metal-free recyclable photocatalyst and CTAB as surfactant in water. Control experiments and the ESR results indicated that superoxide radicals and valence band holes played an important role in the reaction. Further isotope experiments suggested both a beta-scission/HAT pathway and an oxidation/hydrolysis/dehydration pathway for the reaction, which is different from previous reports. The semiconductor/micellar catalyst system can be recycled at least 10 times without a significant reduction in activity. Furthermore, this reaction could be carried out under solar light irradiation and was applicable to large-scale reactions with similar results.

Many natural or synthetic chalcones have potential anti-tumor activity. Here, we synthesized two series of chalcone analogues containing a thieno [2,3-cflpyrimidin-2-y] group and evaluated for their cytotoxic activity towards cultured human lung cancer A549 and colorectal HCT-116 cells. Among them, compound 8d was the most cytotoxic against HCT-116 cells, with an IC50 value of 2.65 mu M. Analyses of the phenotypic changes induced by this compound found a dose-dependent accumulation of HCT-116 cells in sub-G1 phase, indicating that compound 8d might induce apoptosis. Furthermore, we found that 8d triggered mitochondrial membrane potential depolarization, promoted reactive oxygen species formation in HCT-116 cells, and increased the percentage of early and late apoptotic cells. Finally, immunoblotting indicated that 8d increased PARP-1 and caspases 3, 7 and 9 cleavage. These data suggest that compound 8d induces apoptosis via the mitochondrial death pathway.

An amino-functionalized silica-coated Fe3O4 nanocomposite (Fe3O4@SiO2/APTS) was synthesized. The Fe3O4@SiO2 microspheres possessed a well-defined core-shell structure, uniform sizes and high magnetization. An immobilized ruthenium nanoparticle catalyst (Fe3O4@SiO2/APTS/Ru) was obtained after coordination and reduction of Ru3+ on the Fe3O4@SiO2/APTS nanocomposite. The Ru nanoparticles were not only ultra-small with nearly monodisperse sizes but also had strong affinity with the surface of Fe3O4@SiO2/APTS. The obtained catalyst exhibited excellent catalytic performance for the hydrogenation of a variety of aromatic nitro compounds, even at room temperature. Moreover, Fe3O4@SiO2/APTS/Ru was easily recovered using a magnetic field and directly reused for at least five cycles without significant loss of its activity.

An efficient and low-cost supported Pt catalyst for hydrogenation of niroarenes was prepared with colloid Pt precursors and alpha-Fe2O3 as a support. The catalyst with Pt content as low as 0.2 wt% exhibits high activities, chemoselectivities and stability in the hydrogenation of nitrobenzene and a variety of niroarenes. The conversion of nitrobenzene can reach 3170 mol(conv) h(-1) mol(pt)(-1) under mild conditions (30 degrees C, 5 bar), which is much higher than that of commercial Pt/C catalyst and many reported catalysts under similar reaction conditions. The spatial separation of the active sites for H-2 dissociation and hydrogenation should be responsible for the high chemoselectivity, which decreases the contact possibility between the reducible groups of nitroarenes and Pt nanoparticles. The unique surface properties of alpha-Fe2O3 play an important role in the reaction process. It provides active sites for hydrogen spillover and reactant adsorption, and ultimately completes the hydrogenation of the nitro group on the catalyst surface. A three-component reaction has been developed for the construction of multiaryl-substituted pyrrole derivatives from arylketones, amines, and nitrovinylarenes under metal- free conditions. Hence, homologous 1,2,4-triaryl-substituted pyrrole products were obtained in good to high yields. Furthermore, 2,3,5-triaryl-substituted pyrroles were selectively formed in the absence of nitrovinylarenes. A general and efficient boronic acid catalyzed Friedel-Crafts alkylation of arenes with benzylic alcohols was previously developed for the construction of unsymmetrical diarylmethane products (X. Mo, J. Yakiwchuk, J. Dansereau, J. A. McCubbin and D. G. Hall, J. Am. Chem. Soc., 2015, 137, 9694). Highly electron-deficient benzylic alcohols, however, were ineffective coupling partners due to the increased difficulty of C-O bond ionization. Herein, we report the use of perfluoropinacol as an effective co-catalyst to improve the reactivity of a boronic acid catalyst in the Friedel-Crafts benzylations of electronically deactivated primary and secondary benzylic alcohols. According to spectroscopic studies, it is believed that perfluoropinacol condenses with the arylboronic acid catalyst to form a highly electrophilic and Lewis acidic boronic ester. This in situ formed species enables a more facile ionization of the benzylic alcohols likely through a mode of activation promoted by a Lewis acid assisted hydronium Bronsted acid generated from the interactions of the transient boronic ester with hexafluoroisopropanol solvent and water.

The first general palladium-catalyzed alpha-arylation of arylacetonitriles with aryl and heteroaryl sulfonates are reported. Pd(OAc)(2) associated with XPhos serves as the effective catalyst to facilitate this reaction. A broad range of electron-rich, -neutral, -deficient, and sterically hindered aryl/heteroaryl tosylates and mesylates are coupled with arylacetonitriles bearing different substituents to give the corresponding products in good to excellent yields. Catalyst loading down to 0.1 mol-% Pd was achieved, and 22 unprecedented compounds were synthesized from 43 demonstrated examples using this method. Its applicability with the modification of biological phenolic compounds was successfully demonstrated. The Pd/XPhos system catalyzed the alpha-arylation and followed by alkylation in one-pot sequential conditions, resulting in the direct synthesis of compounds containing quaternary center- and deuterium-containing compounds in good to excellent yields.

In the present work, the catalytic potential of natural organo-catalyst thiamine hydrochloride is explored for the synthesis of 1,2,4-triazolidine-3-thiones in aqueous medium. The reaction of thiosemicarbazides with various substrates viz aryl/ heteroaryl aldehydes, isatins, cyclic and aromatic ketones to obtain corresponding 1,2,4-triazolidine-3-thiones have been successfully demonstrated for the first time. The method proposed in this work addresses the green chemistry principles such as biodegradable catalyst, ambient reaction condition, shorter reaction time, high yield of the products, recyclability of catalyst and high atom economy.

A [3+1+1+1] cycloaddition was developed among 2-arylpropene, ketone and DMSO in the presence of K2S2O8. 2-arylpropene provides three carbons, ketone offers one carbon, and DMSO as dual carbon donor contributes two carbons to the six-membered carbocycle. It gave the cyclohexene motif and spirocyclohexene skeleton. Four C-C bonds formed in this process. Both propylene and ketone could be well tolerated and give the corresponding cyclohexene or spirocyclohexene motif in useful yields. A series of 2-phenyl-7-(aryl)pyrazolo [1,5-a]pyrimidine-3-carbonitriles 11a-j and 2-phenyl-7-(aryl)pyrazolo[1,5-a]pyrimidine-3,6-dicarbonitriles 16a-c was synthesized by the reaction of 5-amino-3-phenyl-1H-pyrazole-4-carbonitrile (5) with 3-(dimethylamino)-1-arylprop 2 en 1 ones 6a-j or 2-aryl-3-(dimethylamino)acrylonitriles 12a-c, respectively. In addition, 7-amino-5-oxo-2-phenyl-4,5-dihydropyrazolo [1,5-a]pyrimidine-3-carbonitrile (22) was prepared from the reaction of compound 5 with ethyl cyanoacetate. The anticancer activity of the newly synthesized compounds against Huh-7, HeLa, MCF-7 and MDA-MB231 cell lines showed moderate activity of compound 11 f as anti-proliferative agent against Huh-7 cell line with IC50 = 6.3 mu M when compared with doxorubicin (IC50, = 3.2 mu M). On the other hand, compound 16b revealed potent anti-proliferative activity against HeLa cell line with IC50, = 7.8 mu M when compared with doxorubicin (IC50, = 8.1 mu M). Also compound 11i exhibited a promising anti-proliferative activity against MCF-7 cell line (IC50, = 3.0 mu M) whereas IC50 of doxorubicin = 5.9 mu M, finally compounds 11i and 16b have potent activity as anti-proliferative agents against MDA-MB231 cell line with IC50, = 4.32 and 5.74 mu M, respectively when compared with doxorubicin (IC50 = 6.0 mu M).
