A pathway for the catalytic hydrosilylation of carbonyl substrates with M(C6F5)(3) (M = B, Al and Ga) was calculated by DFT (B3PW91-D3) and it was shown that in the case of the Al reagent, the carbonyl substrate binds irreversibly and inhibits catalysis by generating a stable carbonyl adduct. In contrast, the reduced electrophilicity of B(C6F5)(3) disfavors the binding of the carbonyl substrate and increases the concentration of an activated silane adduct which is the species responsible for catalytic turnover. A similar mechanism was found for both cationic and neutral Re(III) species. Further, it was shown by tuning the electrophilicity of the rhenium catalysts, conditions can be found that would enable the catalytic hydrosilylation of ketone and nitrile substrates that were unreactive in previously reported systems. Thus the mechanisms proposed in this work, lay the foundation for the design of new catalytic systems.

In this contribution, a protocol was established for the selective catalytic hydrogenation of nitroarenes to the correspondingN-arylhydroxylamines. The reduction of 1-(4-chlorophenyl)-3-((2-nitrobenzyl)oxy)-1H-pyrazole, an intermediate in the synthesis of the antifungal reagent pyraclostrobin that includes carbon-chlorine bonds, benzyl groups, carbon-carbon double bonds and other structures that are easily reduced, was chosen as the model reaction for catalyst evaluation and condition optimization. Extensive passivant evaluation showed that RANEY (R)-nickel treated with ammonia/DMSO (1 : 10, v/v) afforded the optimal result, especially with a particle size of 400-500 mesh. To combine the modified catalyst with continuous-flow reaction technology, the reaction was conducted at room temperature, rendering the desired product with a conversion rate of 99.4% and a selectivity of 99.8%. The regeneration of catalytic activity was also studied, and an in-column strategy was developed by pumping the passivate liquid overnight. Finally, the generality of the method was explored, and 7 substrates were developed, most of which showed a good conversion rate and selectivity, indicating that the method has a certain degree of generality.

Background: Diabetes mellitus is the third-largest non-communicable chronic disease worldwide. There are many effective drugs, but the long-term use of these clinical drugs may cause various side effects. Therefore, it is urgent to develop new antidiabetic molecules with higher efficacy and lower toxicity. Methods: Fifteen new 3-aryl-1-(5-methylisoxazol-3-ylamino)-1-(4-nitrophenyl)propan-1-one were synthesized directly through the Mannich reaction of 4-nitroacetophenone, 3-amino-5-methylisoxazole and aromatic aldehydes catalyzed by concentrated hydrochloric acid. The molecular structures of the products were fully characterized by H-1 NMR, C-13 NMR, ESI MS and HRMS. The peroxisome proliferator-activated receptor (PPAR) response element and alpha-glucosidase inhibitory activity of these compounds were evaluated in vitro. Molecular docking, molecular physical parameters calculation, and molecular toxicity prediction were performed to analyze the structure-activity relationship and evaluate the druggability of these compounds theoretically. Results: All compounds exhibited weak antidiabetic activities, but compound 15 showed promising as a high performance, dual-target antidiabetic lead compound with peroxisome proliferator-activated receptor (PPAR) response element relative agonist activity of 99.55% at 27.2 nmol.ml(-1) and alpha-glucosidase inhibitory activity of 35.21% at 13.6 nmol.mL(-1). All compounds obtained may have no cardiotoxicity, no acute toxicity, no carcinogenic, and within safe range of mutagenic risk. Conclusion: This study identified a potential PPAR lead molecule and presented an unusual strategy for antidiabetic drug development.
