A ReCl(CO)(5)/MeC(CH2PPh2)(3) (L2) system was developed for the C-methylation reactions utilizing methanol and base, following the borrowing hydrogen strategy. Diverse ketones, indoles, and arylacetonitriles underwent mono- and dimethylation selectively up to 99% yield. Remarkably, tandem multiple methylations were also achieved by employing this catalytic system.

This work presents three mononuclear Ru(ii) complexes of tridentate phosphine-carboxamide based ligands providing a NNP coordination environment. The octahedral Ru(ii) ion shows additional coordination with co-ligands; CO, Cl and CH3OH. All three Ru(ii) complexes were thoroughly characterized including their crystal structures. These Ru(ii) complexes were utilized as catalysts for the transfer hydrogenation of assorted carbonyl compounds, including some challenging biologically relevant substrates, using isopropanol as the hydrogen source. The binding studies illustrated the coordination of the isopropoxide ion by replacing a Ru-ligated chloride ion followed by the generation of the Ru-H intermediate that was isolated and characterized and was found to be involved in the catalysis.

Chiral, cationic NHC-iridium complexes are introduced as catalysts for the intramolecular hydroamination reaction of unactivated aminoalkenes. The catalysts show high activity in the construction of a range of 5- and 6-membered N-heterocycles, which are accessed in excellent optical purity, with various functional groups being tolerated with this system. A major deactivation pathway is presented and eliminated by using alternative reaction conditions. A detailed experimental and computational study on the reaction mechanism is performed providing valuable insights into the mode of action of the catalytic system and pointing to future modifications to be made for this catalytic platform.

Biocatalytic reduction is a very important field of research in synthetic organic chemistry. Herein, three different Lactic Acid Bacteria (LAB) strains were evaluated for their bioreduction potential using acetophenone as a model substrate. Among these strains, Lactobacillus kefiri P2 strain was determined as the best asymmetric reduction biocatalyst. Reaction optimization parameters such as reaction time, temperature, agitation speed and pH were systematically optimized using Lactobacillus kefiri P2 strain and model substrate acetophenone. Under these optimized reaction conditions, secondary chiral alcohols were obtained by bioreduction of various prochiral ketones with results up to 99% enantiomeric excess. In addition, the steric and electronic effects of substituents on enantioselectivity and conversion were evaluated. It has been shown that Lactobacillus kefiri P2 biocatalyst was an effective catalyst for asymmetric reduction. This method provides an environmentally friendly method for the synthesis of optically pure alcohols and an alternative approach to chemical catalysts.

The direct and efficient oxidation of tertiary benzylic C-H bonds to alcohols with O-2 was accomplished in the presence of metalloporphyrins as catalysts under solvent-free and additive-free conditions. Based on effective inhibition on the unselective autoxidation and deep oxidation, systematical investigation on the effects of porphyrin ligands and metal centers, and apparent kinetics study, the oxidation system employing porphyrin manganese(II) (T(2,3,6-triCl)PPMn) with bulkier substituents as catalyst, was regarded as the most promising and efficient one. For the typical substrate, the conversion of cumene could reach up to 57.6% with the selectivity of 70.5% toward alcohol, both of them being higher than the current documents under similar conditions. The superiority of T(2,3,6-triCl)PPMn was mainly attributed to its bulkier substituent groups preventing metalloporphyrins from oxidative degradation, its planar structure favoring the interaction between central metal with reactants, and the high efficiency of Mn(II) in the catalytic transformation of hydroperoxides to alcohols.

Fourteen N-[{(substituted-phenylthiazol-2-yl)-3-aryl-1H-pyrazol-4-yl}methylene]-5-substituted-thiazol-2-amine (5a-n) analogs were synthesized by the reaction of 3-aryl-1-(thiazol-2-yl)-1H-pyrazole-4-carbaldehyde and substituted thiazole amines. The structures of prepared compounds were delineated by elemental analysis, FT-IR and H-1 NMR spectra. These analogs were scrutinized for in vitro antiinfective and cytotoxic activities. Some thaizole clubbed pyrazole derivatives were assessed for their cytological changes in germ cells of Capra hircus by using histomorphological analysis, fluorescence assay and apoptosis quantification. Compound 51 having 4-NO2 substituent induced the significant apoptosis in tested cells of Capra hircus. The results revealed that compounds 5c, 5e, 5k, and 51 have commendable antibacterial activity within MIC range of 62.5-250 mu g/ml. Compound 5c emerged as a potent antimalarial compound by exhibiting IC50 value of 0.23 mu g/ml and compound 5j induced paralysis of Pherentima posthuma at 8.6 +/- 1.94 min and death at 20 +/- 5.04 min, respectively. Compound 5j revealed an excellent cytotoxicity at IC50 value of 30.7 and < 10 mu g/ml against MCF-7 and HeLa cells, respectively. Described here is the design and discovery of donor-acceptor-type platinum catalysts that are highly effective in both hydroalkoxylation and hydroamination of unactivated alkenes over a broad range of substrates under mild conditions. A number of alkene substitution patterns are accommodated, including tri-substituted, 1,1-disubstituted, (E)-disubstituted, (Z)-disubstituted and even mono-substituted double bonds. Detailed mechanistic investigations suggest a plausible pathway that includes an unexpected dissociation/re-association of the electron-deficient ligand to form an alkene-bound donor-acceptor-type intermediate. These mechanistic studies help understand the origins of the high reactivity exhibited by the catalytic system, and provide a foundation for the rational design of chiral catalysts towards asymmetric hydrofunctionalization reactions. The direct addition of an O-H (hydroalkoxylation) or an N-H (hydroamination) bond across an alkene is a useful strategy to access biologically relevant heterocycles. Here, the authors report donor-acceptor-type platinum catalysts that are effective for both hydroalkoxylation and hydroamination of unactivated alkenes, with broad substrate scope and use of mild conditions.
