This is the first report of the use of multicomponent reactions and subsequent cascades to access complex, unprecedented organocesium architectures. Moreover, this article also highlights the first use of mild cesium carbonate as a cesium source for the construction of cesium organometallic scaffolds. Relativistic DFT calculations provide an insight into the electronic structure of the reported compounds. Stereo-selective bio-reduction of acetophenone and its derivatives by soil fungal isolates

Microbial asymmetric reduction of ketone is an efficient tool for the synthesis of chiral alcohols. Many reports are available for the bio-reduction of acetophenone and its derivatives, since it is widely used model substrate. This research focus on exploring the soil fungal isolates for their ability towards the reduction of acetophenone and its derivatives to their corresponding chiral alcohols using growing cells instead of resting cells. Bio-reduction of acetophenone, 4-fluoro acetophenone, and 4-chloro acetophenone were carried out using different fungal cultures isolated from soil. All the isolates exhibited a good reduction capability when grown in an optimal condition. Among the screened fungal cultures, Penicillium sp. and Aspergillus sp. showed significant bioconversion with varying enantio-selectivity. However, the Penicillium sp. has showed maximum ability of bio-reduction. The best performing isolate was characterized using internal transcribed spacer (ITS) region and found to be Penicillium rubens VIT SS1, which showed higher conversion and selectivity more than 90% towards acetophenone and its derivatives. The reaction conditions such as pH, temperature and media were evaluated for the bio-reduction of acetophenone using Penicillium rubens VIT SS1. The substrate loading was increased from 0.5g/L to 6g/L at shake flask level using the optimized condition pH 5±0.5 and temperature 25±2C. This study revealed huge potential of fungal cultures for the synthesis of many aromatic chiral alcohols in a simpler, novel and cost effective manner.

Structural studies and characterization of 3-formylchromone and products of its reactions with chosen primary aromatic amines

The crystal structure and FTIR spectrum of 3-formylchromone (0) have been analysed. A new organic ligands, 3-(anilinomethylene)-2-methoxychroman-4-one (1) and 3-(1?,4?-benzodioxan-6?-aminomethylene)-2-methoxychroman- 4-one (2) were synthesized. The products of reactions between 3-formylchromone and aniline and 6-amino-1,4,-benzodioxane were studied by elemental analysis, FTIR spectroscopy, thermal analysis and X-ray crystallography. It has been found that these reactions allow to obtain derivatives of 4-chromanones from 3-formylchromone by the mechanism of nucleophilic addition, accompanied by the opening of the pyrone ring. The experimental results indicate that compounds (1) and (2) are thermal stable up to 90 and 100 C, respectively. The FTIR spectra and X-ray analysis showed the present of methoxy-group in these compounds.

In this paper, quaternary 8-(1-acylethene-1-yl)-13-methylcoptisine chlorides targeting thioredoxin reductases (TrxRs) were designed to test the growth inhibitory activity against human cancer cell lines and the effect on viability of the normal intestinal epithelial cell-6 (IEC-6) in vitro and to evaluate structure-activity relationship (SAR). The introduced alpha beta-unsaturated ketone groups at C-8 consisting of n-alkanoyls possessing five to ten carbons or aroyls or cyclohexylcarbonyl increased the tested activity against the target cancer cell lines. By and large, this type of improvement was increasingly graced by the elongation of the aliphatic chain of the n-alkanoyls in the range of less than ten carbon atoms. The relatively more polar 1-acylethene-1-yls displayed no effect on improving the activity. All the explored aroyls showed significant effect on improving the activity of the target compounds against the tested cancer cell lines with no SAR being observed. The findings of this study suggested that oil/water partition coefficient of the test compounds was one of the key factors impacting the target activity against the tested cancer cell lines. At the concentration of 10 mumol/L, except for the compounds with n-alkanoyls possessing seven or more carbons or with alpha-naphthoyl, none of the other compounds displayed obvious cytotoxicity on normal IEC-6 cell when co-incubated. The survival rate of IEC-6 cell ranged from 75% to 100% for the noncytotoxic compounds.
