Stabilization of ketone and aldehyde enols by formation of hydrogen bonds to phosphazene enolates and their aldol products

Solution properties of enolates generated using the phosphazene (Schwesinger) base P4-tBu were investigated by NMR spectroscopy. With a full equivalent of base the benzyl ketones 1a and 1b, the acetophenone 2, the arylacetaldehyde 1c, and the methyl arylacetate 1d formed the expected “naked” (P4H+) enolates 3 and 7. However, at a half-equivalent of base the ketones 1a and 1b as well as the aldehyde 1c formed solutions of stable hydrogen-bonded dimeric (enol-enolate) structures (4). The acetophenone 2, on the other hand, forms only traces of the H-bonded dimer 8 during deprotonation of 2. The thermodynamic product was the isomeric self-aldol condensation product 12. The mechanism of this condensation was elucidated by low temperature rapid-injection (RI) NMR spectroscopy. Solutions of 8 stable enough for NMR characterization could be transiently generated by semiprotonation of the enolate 7 with HCl¡¤OEt2 at -130 C using RINMR. The ester enolate 1d gave no trace of 4d even on a time scale as short as a few seconds at -130 C either during the semideprotonation of 1d, or during semiprotonation of the enolate 3d. Long-lived solutions of the enols derived from 1a, 1b, 1c, and 2 (but not 1d) could be produced by full protonation of the phosphazene enolates with HCl¡¤OEt2 at low temperature. Copyright

Directed evolution of P450 BM3 towards functionalization of aromatic O-heterocycles

The O-heterocycles, benzo-1,4-dioxane, phthalan, isochroman, 2,3-dihydrobenzofuran, benzofuran, and dibenzofuran are important building blocks with considerable medical application for the production of pharmaceuticals. Cytochrome P450 monooxygenase (P450) Bacillus megaterium 3 (BM3) wild type (WT) from Bacillus megaterium has low to no conversion of the six O-heterocycles. Screening of in-house libraries for active variants yielded P450 BM3 CM1 (R255P/P329H), which was subjected to directed evolution and site saturation mutagenesis of four positions. The latter led to the identification of position R255, which when introduced in the P450 BM3 WT, outperformed all other variants. The initial oxidation rate of nicotinamide adenine dinucleotide phosphate (NADPH) consumption increased ?140-fold (WT: 8.3 ¡À 1.3 min-1; R255L: 1168 ¡À 163 min-1), total turnover number (TTN) increased ?21-fold (WT: 40 ¡À 3; R255L: 860 ¡À 15), and coupling efficiency, ?2.9-fold (WT: 8.8 ¡À 0.1%; R255L: 25.7 ¡À 1.0%). Computational analysis showed that substitution R255L (distant from the heme-cofactor) does not have the salt bridge formed with D217 in WT, which introduces flexibility into the I-helix and leads to a heme rearrangement allowing for efficient hydroxylation.

Fatty acid biosynthesis is essential for bacterial survival. ss-Ketoacyl-acyl carrier protein (ACP) synthase III (FabH), is a particularly attractive antibacterial target, since it is central to the initiation of fatty acid biosynthesis. Three series of 21 cinnamaldehyde acylhydrazone derivatives, A3-9, B3-9, and C3-9, were synthesized and evaluated for FabH-inhibitory activity. Compound B6 showed the most potent biological activity against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Bacillus subtilis (minimum inhibitory concentrations (MICs) values: 1.56-3.13mug/mL) and was comparable with the positive control. Docking simulation by positioning compound B6 in the FabH structure active site was performed to explore the possible binding model.

Furthermore, 62 decreased LDL-C without elevating the plasma TG level and significantly suppressed the lipid-accumulation area in the aortic arch in a Bio F1B hamster fed a diet high in fat and cholesterol. We demonstrated that our LXRbeta agonist would be potentially useful as a hypolipidemic and anti-atherosclerotic agent. In this manuscript, we report the design, synthesis and pharmacology of 1,3-bistrifluoromethylcarbinol derivatives.

A hypercrosslinked polymer (HCP)-immobilized ruthenium catalyst (HCP?PPh3?Ru) was prepared from benzene and triphenylphosphine (PPh3) through a one-step external cross-linking reaction. The structure and composition of the catalyst were fully characterized by many physicochemical methods. HCP?PPh3?Ru was found to possess high Brunauer?Emmett?Teller (BET) surface area, large pore volume, and low skeletal density. Moreover, the Ru catalyst was also featured some advantages, such as low synthetic cost and good chemical and thermal stability. HCP?PPh3?Ru was then used as a recoverable and effective catalyst for the synthesis of 2,4-diaryl-substituted pyridines from acetophenones, ammonium acetate (NH4OAc) and dimethylformamide (DMF), and the cycloaddition reaction of diazodicarbonyl compounds with olefins to afford dihydrofurans. In addition, a combination of HCP?PPh3?Ru and Amberlyst-15 enabled us to establish a simple protocol for the direct synthesis of carbazole derivatives from diazodicarbonyl compound, alkyl vinyl ether and indole. In these selected reactions, the catalyst was easily recovered and reused several times without significant activity loss. (Figure presented.).

Synthesis and cytotoxic activities of a series of novel N -methyl-bisindolylmaleimide amide derivatives

A novel series of N-methyl-bisindolylmaleimides were synthesized and evaluated for their inhibitory activities against nine tumor cell lines. Exploring the Existing Drug Space for Novel pTyr Mimetic and SHP2 Inhibitors

Protein tyrosine phosphatases (PTPs) are potential therapeutic targets for many diseases. Unfortunately, despite considerable drug discovery efforts devoted to PTPs, obtaining selective and cell permeable PTP inhibitors remains highly challenging. We describe a strategy to explore the existing drug space for previously unknown PTP inhibitory activities. This led to the discovery of cefsulodin as an inhibitor of SHP2, an oncogenic phosphatase in the PTP family. Crystal structure analysis of SHP2 interaction with cefsulodin identified sulfophenyl acetic amide (SPAA) as a novel phosphotyrosine (pTyr) mimetic. A structure-guided and SPAA fragment-based focused library approach produced several potent and selective SHP2 inhibitors. Notably, these inhibitors blocked SHP2-mediated signaling events and proliferation in several cancer cell lines. Thus, SPAA may serve as a new platform for developing chemical probes for other PTPs.
