Phenylamine phenanthroimidazole based bipolar compounds with donor-acceptor (D-A) architecture namely, 4-(1-(2,3-dihydrobenzo[b][1,4]dioxin-5-yl)-6,9-di(pyren-4-yl)-1H-phenanthro[9,10-d]imidazol-2-yl)-N,N-diphenylaniline (DDPPPA) and 4?-(1-(2,3-dihydrobenzo[b][1,4]dioxin-5-yl)-6,9-di(pyren-4-yl)-1H-phenanthro[9,10-d]imidazol-2-yl)-N,N-diphenyl-[1,1?-biphenyl]-4-amine (DDPBA) have been synthesized with highly fluorescent pyrene moieties at C6- and C9-positions. The C6 and C9 modification enhanced the thermal, photochemical and electroluminescent properties. Both molecules were employed as blue emitters in non doped organic light emitting devices (OLEDs) and show high performances due to hybridized local and charge-transfer properties. An OLED with DDPPPA/DDPBA emissive layer shows deep-blue emission with maximum external quantum efficiency (etaex), current efficiency (etac) and power efficiency (etap) of 5.7/6.0%, 10.5/12.0 cd A-1 and 8.3/9.2 lm W-1, respectively. Both devices show high singlet exciton utilizing efficiency (etas) of DDPPPA-31.33% and DDPBA-35.29%. The doped device m-MTDATA:DDPPPA/m-MTDATA:DDPBA shows maximum efficiencies of etac -7.4/8.23 cd A-1; etap -5.8/6.13 lm W-1; etaex -4.72/5.63% (5 wt%):etac -8.36/9.15 cd A-1; etap -6.32/6.65 lm W-1; etaex -4.86/5.45% (10 wt%):etac -9.58/10.02 cd A-1; etap -7.8/8.25 lm W-1; etaex -5.96/6.25% (20 wt%). The doped device based on TAPC host TAPC:DDPPPA/TAPC:DDPBA exhibits maximum efficiencies of etac -9.60/10.03 cd A-1; etap -7.81/8.26 lm W-1; etaex -5.96/6.25% (20 wt%).

Design, synthesis and biological evaluation of novel human monoamine oxidase B inhibitors based on a fragment in an X-ray crystal structure

Herein we report our efforts of developing reversible selective hMAO-B inhibitors based on isatin, a fragment in an X-ray crystal structure. Five different scaffolds were designed and many compounds were synthesized. Among them, compound A3 demonstrated very high potency and isoform selectivity against hMAO-B, 11 and 13 times more potent (IC50 = 3 nM) and 23.64 and 6.8 times more selective than the marked drugs, selegiline and safinamide. However, the endeavors to modify the polar 3-one group of isatin, that is in a hydrophobic environment in the binding site of hMAO-B, to small nonpolar hydrophobic groups did not bring about improved hMAO-B inhibitors, which may challenge our understanding of molecular interactions and molecular recognition in biological systems.

A novel series of potent and selective non-peptide neuropeptide Y (NPY) Y1 receptor antagonists, having benzazepine nuclei, have been designed, synthesized, and evaluated for activity. Chemical modification of the R1 and R3 substituents in structure 1 (Chart 1) yields several compounds that show high affinity for the Y1 receptor (K(i) values of less than 10 nM). SAR studies revealed that introduction of an isopropylurea group at R1 and a 3- (benzo-condensed-urea) group, 3-(fluorophenylurea) group, or a 3-(N-(4- hydroxyphenyl)guanidine) group at R3 in structure 1 afforded potent and subtype-selective NPY Y1 receptor antagonists. 3-(3-(Benzothiazol-6- yl)ureido)-1-N-(3-(N’-(3-isopropylureido))benzyl)-2,3,4,5-tetrahydro-1H-1- benzazepin-2-one (21), which was one of the most potent derivatives, competitively inhibited specific [125I]peptide YY (PYY) binding to Y1 receptors in human neuroblastoma SK-N-MC cells (K(i) = 5.1 nM). 21 not only inhibited the Y1 receptor-mediated increase in cytosolic free Ca2+ concentration in SK-N-MC cells but also antagonized the Y1 receptor-mediated inhibitory effect of peptide YY on gastrin-induced histamine release in rat enterochromaffin-like cells. 21 showed no significant affinity in 17 receptor binding assays including Y2, Y4, and Y5 receptors.

Catalytic reductions of carbonyl-containing compounds are highly important for the safe, sustainable, and economical production of alcohols. Herein, we report on the efficient transfer hydrogenation of ketones catalyzed by a highly potent Mn(I)?NHC complex. Mn?NHC 1 is practical at metal concentrations as low as 75 ppm, thus approaching loadings more conventionally reserved for noble metal based systems. With these low Mn concentrations, catalyst deactivation is found to be highly temperature dependent and becomes especially prominent at increased reaction temperature. Ultimately, understanding of deactivation pathways could help close the activity/stability-gap with Ru and Ir catalysts towards the practical implementation of sustainable earth-abundant Mn-complexes.
