General procedure: 1a-j (0.0299 – 0.0532 g, 0.299 mmol, 1 eq.), 2a-d (0.0166 – 0.228 g, 0.224 mmol, 0.75 eq.), FeCl3 (0.0145 g, 0.0897 mmol, 0.3 eq.), (NH4)2S2O8 (0.1363 g, 0.598 mmol, 2 eq.), and DCE / CCl4 (0.75 mL, 1 / 1) were stirred in Schott culture tubes (H ¡Á diam. 160 mm ¡Á 16 mm) at 120 C for 3 h. Further, after adding a solution of 2a-d (0.055 – 0.0763, 0.0748 mmol, 0.25 eq.) and DCE / CCl4 (0.08 mL, 1 / 1), the reaction mixture was stirred at 120 C for 3 h (3 times at 3-hour intervals). Next it was stirred at 120 C for 12 h. The yields of 3a-f,a-d were determined by 1H NMR (CDCl3) after the filtration of the reaction mass through a thin pad (0.4 – 0.5 cm) of silica gel (0.015 – 0.040 mm) using 20 mL of DCM and evaporation of solvent in vacuum. Products 3c,b and 3d,a-d were isolated by gradient flash chromatography (eluent: petroleum ether / ethyl acetate).

General procedure: A mixture of compound 5 (0.13 mmol), the appropriate acid (0.17 mmol), HOBt (0.17 mmol), and EDCI (0.19 mmol) in dry DMF was cooled to 0 ¡ãC under nitrogen atmosphere. To the reaction mixture, triethylamine (0.16 mmol) was added at 0 ¡ãC. The mixture was then stirred at 90 ¡ãC for 12 h. The reaction mixture was cooled and then partitioned between water and ethyl acetate and the organic layer was separated. The aqueous layer was then extracted with ethyl acetate and the combined organic extracts were washed with brine and dried over anhydrous Na2SO4. After evaporation of the organic solvent, the residue was purified by column chromatography to afford the corresponding compounds 1a-l.

1,4-Benzodioxan-5-carboxylic acid (120 g) was added in portions at 40¡ã C. to a stirred mixture of hydroxylamine hydrochloride (52 g) and polyphosphoric acid (380 g), then the mixture was heated to an internal temperature of 120¡ã C. The source of heat was removed, and the mixture was stirred vigorously until frothing subsided, then it was stirred at 165¡ã C. for 90 minutes, cooled to 80¡ã C., added to an excess of ice-water, and basified by the addition of 5M aqueous sodium hydroxide solution. The product was extracted into ethyl acetate (6*500 ml), the extracts were dried (MgSO4), and the solvent removed in vacuo. The residue was dissolved in ethyl acetate (800 ml), and the cloudy solution was filtered then saturated with hydrogen chloride. The resulting solid was collected by filtration, washed with ethyl acetate, and dried in vacuo at ambient temperature to give 1,4-benzodioxan-5-amine monohydrochloride as a buff solid (65.9 g).

General procedure: The initial carbonyl compound (50mmol) was dissolved/suspended in ethanol (50mL) and magnetically stirred with thiosemicarbazide (50mmol) and catalytic amounts of acetic acid for 8-24hat room temperature. The obtained thiosemicarbazone was filtered, washed with the appropriate solvent (n-hexane, petroleum ether or diethyl ether) and dried under vacuum. Ethyl-bromoacetate (50mmol) was added to a suspension of the intermediate thiosemicarbazone (50mmol) and sodium acetate (50mmol) in methanol (50mL) and the resulting mixture stirred at room temperature for 24-48h. The obtained crude 4-thiazolidinone was poured on ice, filtered or extracted with chloroform (3¡Á30mL) and purified by chromatography (SiO2, ethyl acetate/n-hexane 1/2) to give compounds 1a-58a in high yields. The product (50mmol) was dissolved/suspended in 50mL of anhydrous acetone in the presence of anhydrous potassium carbonate (50mmol), and reacted with equimolar amounts of benzyl bromide for 24-48h. The mixture was poured on ice, filtered or extracted with chloroform (3¡Á50mL) and purified by column chromatography (SiO2, ethyl acetate/n-hexane 1/3) in order to obtain compounds 1b-58b in high yield as previously reported [25,26].
