EXAMPLE 22 Preparation of (-)-N-[[3-[4-[4-[(2,3-Dihydro-1,4-benzodioxin-2-yl)carbonyl]-1-piperazinyl]-3-fluorophenyl]-4,5-dihydro-5-isoxazolyl]methyl]acetamide. To a flame dried flask containing (-)-N-[[4,5-dihydro-3-[3-fluoro-4-(1-piperazinyl)phenyl]-5-isoxazolyl]methyl]acetamide (Step 1, Example 21) (200 mg), 1,4-benzodioxan-2-carboxylic acid (135 mg), DMAP (9 mg) in pyridine (7 mL) is added EDC (144 mg) at ambient temperature and stirred for 48 hours. The reaction is diluted with CH2Cl2 (25 mL) and washed with H2O (2 X 50 mL) and saline (50 mL). The organic phase is dried over sodium sulfate, concentrated in vacuo and chromatographed on silica gel (230-400 mesh), eluding with chloroform/methanol (99/1). The appropriate fractions are combined (Rf= 0.29,TLC, chloroform/methanol, 95/5), concentrated to give the title compound, mp 158-161 ¡ãC, [alpha]25D -43¡ã (CHCl3)., 3663-80-7

Step 1..Preparation of 3-((2,3-dihydrobenzo[b][1 ,4]dioxin-6-yl)methyl)-2-imidazolidin-4- one; A suspension of (2,3-Dihydro-benzo[1 ,4]dioxin-6-yl)-methylamine (16.5 mmol, 2.73 g) in 1 MNaOH is refluxed for 10 min and cooled to room temperature. Subsequently bis(carboxymethyl) trithiocarbonate (3.74 g, 16.5 mmol) is added and the reaction mixture refluxed overnight to give the precipitate, which is filtered off and is washed with water (3x 20 ml_). The remaining residue is suspended in a mixture of methanol and isopropanol (1 :1 , 25 ml_) and refluxed for 10 minutes. Cooled reaction mixture is filtered and the solid residue dried at lower temperature and recrystallyzed from methanol to give 3.29 g of 3-((2,3- dihydrobenzo[b][1 ,4]dioxin-6-yl)methyl)-imidazolidin-2,4-dione.

Step B: Preparation of 6-nitro-3,4-ethylenedioxybenzoic acid methyl ester. To a solution of 5.0 g. of 3,4-ethylenedioxybenzoic acid methyl ester in 5.0 mls. of glacial acetic acid is added dropwise 5.0 mls. of 70percent nitric acid at 50¡ã-60¡ã C. After addition the reaction mixture is kept at 55¡ã C. for one hour, then cooled and 50 mls. of ice water added. The resulting precipitate is recovered by filtering, water washed and dried to obtain 6-nitro-3,4-ethylenedioxybenzoic acid methyl ester, m.p. 115¡ã-118¡ã C.

General procedure: To a stirred solution of 1 (0.84 mmol) in 5 N NaOH (3 mL) was added 2-heptanone (7.18 mmol) dropwise. The reaction mixture was heated to 60 C and kept for 3 h under stirring conditions. After cooling, the mixture was then extracted with CHCl3/MeOH (10:1, v/v) in a separatory funnel. The organic layer was washed to neutral status using water, dried using anhydrous MgSO4, and filtered. The filtrate was concentrated under reduced pressure to yield a crude intermediate. The crude intermediate was dissolved in anhydrous tetrahydrofuran (8 mL), followed by adding HOAc (1.0 mL) and formaldehyde (15.06 mmol) to the solution. The reaction mixture was kept refluxing for 3 h. After the reaction was halted and the reaction mixture cooled, evaporation under reduced pressure was done to remove the solvent from the reaction mixture. The residue was acidified using aqueous 2 N HCl (2 mL), and the solution was stirred at room temperature for 1 h, and extracted with CHCl3/MeOH (10:1, v/v) in a separatory funnel. The organic layer was dried using anhydrous MgSO4, filtered, and concentrated under reduced pressure to yield the crude product. The crude product was purified using silica gel CC (elutated with CHCl3:MeOH, 20:1, v/v) to yield 3e (97 mg, 23.3% yield) as a yellow solid.
